Durch Mutagenisierung erfolgte im Helicobacter-Stamm 26695 eine Ausschaltung der Gene für die Carboanhydrasen CahA und IcfA. Während der Verlust einer Carboanhydrase zu keinem Wachstumsnachteil führte erwies sich der Verlust beider Enzyme für das Bakterium als letal. In nachfolgenden Wachstumsversuchen wurde die Bedeutung der Carboanhydrasen für H. pylori unter verschiedenen Bedingungen weiter untersucht, wobei insbesondere der Zusammenhang mit Substraten und Produkten der Urease-Reaktion bedeutsam erschien. Die Ergebnisse weisen deutlich auf einen Einfluß der Enzyme auf den wichtigsten Pathogenitätsfaktor, die Urease hin, deren Aktivität durch die Mutation signifikant erniedrigt wurde. Insbesondere scheint eine Funktion in der Aktivierung der Urease-Präproteine zu bestehen..
Mutational analysis of the Helicobacter pylori carbonic anhydrase homologs CahA and IcfA showed a significant influence of these enzymes on the urease activity of strain 26695. The results of this research indicate an important role of the carbonic anhydrases in the activation of urease-pre-proteins of strain 26695. |
